Dear Sir,
I, Muni graduate of ABC would like to apply for the post of Research Officer/Internship in your department. I did my graduation in October 2003, currently I am doing an honorary internship in CDE. If accepted I feel that this experience would be beneficial both for the department and myself.
I have enclosed my Curriculum Vitae in support of my application. If need for further information please contact me on the address below.
Thanking you,

Yours truly,

That's pretty good, Muni. You might think about adding why you feel you will be beneficial to his department. For example, you could mention a strong characteristic of yours (e.g., a strong willingness to learn or good organizational skills), or a success that you had at CDE or in college (e.g., you were given greater responsibility after only two months at CDE). This is not a must, though.

Best of luck!

I agree with Bud except that I was taught to use a comma after Dear Sir and a comma after Yours sincerely. In addition, I was taught to use Yours faithfully rather than Yours sincerely, (which is reserved for business associates with whom you are already on friendly terms.)
