This will be my 5th year.

If I extend my E-2 in June with my current employee (been there for 2 years), am I locked in for 10 more months there? That is to say, what if I signed a new 1-year contract, but in 3 months or 6 months I wanted to go somewhere else in Korea and they were amicable with that? Would I have to leave the country and go home first to change jobs?

You would have to leave the country and get a visa in Japan. If you are 10 months in or finishing up, you can transfer your visa without having to leave the country.
