Does anyone know what the penalties may be, if any, for someone who chooses not to go before their flight to Korea, for whatever reason?

If you have your work visa, you will not be able to get another until your contract expires, i.e. one year.

Theoretically you could be sued for breach of contract.

In realistic terms there are NO consequesnces other than perhaps not being able to get another E2 visa for the next year.

Valkyrian summed it up quite nicely. I when I was in Korea I backed out of a contract at the last minute because i never liked the conditions.
Luckily I did so at the time because my documents were not sent to immigration for processing. The recruiter said they were going to sue me for breaching a contract. However, I called his bluff and nothing happened.

Nothing will happen. Don't worry about it. You'll probably get a nasty email, just forget about it. You may have to wait another year, you may not if you apply to different part of Korea(Immigration really doesn't know what it is doing here). You won't get sued, never happens.

Don't worry. The school will be too busy to find someone new to bother too much about you. It happens all the time.

If you have the visa stamp already in your passport, you are stuck for a year in getting another job.

If you have only received the visa issuance number from the employer and NOT got anything in your passport, you are stuck for 90 days as that is when the number expires in immigration.

If you have only signed a contract and agreed to take the job,(then back out) you are not stuck with anything, and can accept any new job. Unfortunately the employer is up for some new expenses in sorting someone else, and it is far too expensivefor him to sue you in another country.
