When I arrive in Korea, do I need to go to a hospital immediately to get my blood/drug test? Or when do I need to do this?

You need to get it done before you get your Alien registration card. You must get the ARC within 90 days after arrival. You can't get medical insurance until you get your ARC, so you may have to pay more, though technically you are covered under medical from the day you land in Korea on an E2 visa, and must pay into it. So, go get the medical at a higher fee, the once you get your ARC and medical insurance, the hospital will reimburse you money.
