My current contract ends at the end of this week, and I was lucky enough to get a contract for a new school for October 10th. I will likely do the visa transfer sometime this week - how long does it take? As happy as I am to be in Korea for another year (especially considering the offer is really good), I'd like to take a break and go overseas for a bit so I'm a little worried about how long it'll take.

Also, do I need a letter of release from my current school even if I'm finishing my contract and the new job starts 10 days later? My new school says that's the case.

IF you have time on your ARC AFTER your current contract and do the paperwork after your contract and before your expiry you do not need a LOR.

IF you do the paperwork for the transfer during the last week of your current contract (and before your ARC expires) you do not need a LOR.

When you do go to immigration the transfer process takes about 30 minutes.
You need your:
ARC
passport
new contract
application
fees (60k for the transfer and 30k for the visa extension)
copy of the new employer's business registration
letter of guarantee (sponsorship form) from the new employer.

Looks like I won't be able to do it anyway. Because I already extended my visa once (tourist visa, wanted to stick around after the current job in case I couldn't find one in time), it looks like I'll have to do a visa run.

I've sent the necessary documents to my new employer. Will immigration not process the files until I've left the country/cancelled my current visa?

If you are on an E-visa they won't start till it is canceled/expired.
If you are on a B or C visa stamp/visa then yes, they will start and you can do a visa run in about a week when you get your confirmation number.
