Location: I get so little foreign language experience, I must be in Koreatown, Los Angeles.

How do you play 끝말잇기?
My Korean sign language class wanted to play that game,
but they couldn't
because I didn't know how.

I noticed a thread entitled something like "Korean word game."
Is that the same game?

It's the same game, making a word beginning with the last character of the previous word. The name is 끝말잇기, from 잇다 (meaning "to link together").

I corrected my mistake, both in the post and in the title of the thread.

One more question:
What if the last word ends in a vowel?
Should the next word start with an ㅇ?

For instance, if the last word is 고양이,
could the next word be 읽다?

You're only supposed to use nouns, so verbs like 읽다 would be out. Also, the linked syllable has to be exactly the same, so if the word is 고양이 the next word would have to have only 이 as the first syllable. The only exception I can think of is when the syllable begins with ㄹ. In those cases, you can drop the ㄹ or replace it with ㄴ. For example: 화로 > 노력 is OK, and 노력 > 역설 is OK.
