Q1:Can I use then instead:
You finish your homework first, then I will take you to a movie.

Q2:What are the differences between these two sentences?

Yes, you may use "then" in place of "and." However, it changes the type of sentence.

The original sentence with "and" is a case of joining 2 sentences with a conjunction.

When the word "then" is used you form a complex sentence with an independant and dependant clause.
