The main meaning is to (go)(come) closer to something, and in "Step right up, folks.", that is the most likely meaning.

The context is,
The host barked out, "Step right up, folks. Each game has a winner...if you are the first person to ..."

I see.
Of course the person who says "Step right up" doesn't only want you to get closer; getting you closer is the first step in getting you to do something else, usually giving them some of your money.
"step (right) up" has another meaning, which relates to your example too. This meaning is to offer your help or cooperation in order to get something done.
A day before the deadline we still had a lot of work to do but our neighbors stepped right up to help us and we finished the work in time. |
