The second sentence is wrong. It should be 'What does John look like?'

Do you know the difference now?

Are they the same

"What is John like?" asks about his qualities or characteristics. You can say something like John is friendly and funny but he is lazy and dishonest.

"What does John look like?" asks about his appearance. You can say something like He is tall and thin and has dark wavy hair and a round bearded face.
