2.There are an estimated 10,000 acupuncturists in the United States, 6,500 of whom are certified.

Which is correct and why?

Thanks.

An estimated 10,000 is correct - estimated is used as an adjective.

2.an estimated 10,000 acupuncturists

I wonder why 'an' should be used although 'acupuncturists' is plural in the example. I think that 'an' should be removed because 'acupuncturists' is plural, so I think that #1 in the above is more correct than #2. Can you clear my question?

Thanks.

Your question has already been answered. An goes with estimated not with acupuncturists. It's an estimated number or quantity.
