I was taught that "thus" means "in this way." Therefore:
1. Correct as is.
2. Only correct without the "and." If you want "and," change it to: "They diluted the drug, and thus they reduced its effectiveness."
3. Not correct. Change "thus" to "therefore" and it's OK.
4. Same as 3.
