Your sentence is correct and natural, with a correction in spelling - Whom, not whome.
While your sentence is good as is, you could replace 'want' with 'wish' to make it more formal and polite: "Whom do you wish to speak to." It is a sentence likely to be said in the workplace and other formal circumstances.
Be aware, though, that in casual speech we usually don't bother with 'whom.' (At least that's true for Amer. Eng. I'm not sure if it's the same for Brit. Eng.) In informal situations, we'd say "Who do you want to speak to?"
