Could please tell me what does "Are you in the line" mean?

And when can I use it?

You arrive at a theatre and there is a line of people waiting to get in. Some people standing near what you think is the end of the line; they may be waiting to get into the theatre or they may just be standing there having a chat. You don't want to push in ahead of them if they are actually waiting to get into the theatre, so you ask, "Are you in the line?"

If you are in New York City, you may also hear "are you on line?" which means the same thing. I don't know the origin, but they just say it different there.

I tend to use "Are you in line" as opposed to "in the line" myself.
