Is there much of a difference in speed between a memory stick running DDR2 SO-DIMM at 800MHz versus one running at 667MHz?

Also, what is the difference in PC2-6400 memory versus PC2-5300??

I'm running 32-bit Vista and want to upgrade from 2GB to 4GB.

Speed and over clock ability.
Bigger number = more speed, as long as your computer supports it.

Also if youve got 32bit windows itll max out at 3GB of RAM that it can use. So your buying an extra 1GB for nothing, until you get 64bit version going.

pc6400 ram runs at 800mhz. pc5300 ram runs at 667mhz. I f you run mismatched sticks of ram, the faster one will run at the speed of the slower one, kind of a waste if you are doing this.

These guys are right, but just to lay it out a little more simply:

1. It's true your computer will not be able to see all 4GB of RAM. However, I believe it's actually 3.5GB that you can use. If you need more RAM I wouldn't consider the upgrade a waste.

2. If you have some "fast" RAM and some "slow" RAM it will all run at the "slow" speed.

And the questions you actually asked:
3. You probably won't notice a difference between 667 and 800mhz RAM.

4. The difference (as chev said) is that PC2-6400 runs at 800mhz and PC2-5300 runs at 667mhz.

So,
What I would do if I were you is grab 2GB of whatever is cheapest. The 800 speed is kind of the standard nowadays so it might actually be cheaper.
