For about 5 days of relaxation, good value, and beautiful beaches, which of the two do you recommend?

I visited Koh Samet (in 2001) for a night or two, but not the one in Vietnam. It was nothing special but I liked the boat ride and that tropical green water. It was quiet and relaxing though and the beach was OK, not many people there. You could go out into the water for a swim. I had a nice time also because I was with a girl I liked.

Do the online search. I'm sure I've heard good things about the place in Vietnam. Of course you have other options too unless you are determined to choose one or the other.

For just 5 days, matters not much I reckon.

I haven't been to Koh Samet, so maybe I shouldn't have voted, but I really liked Phu Quoc.
My gf and I spent a week there in August and had a great time. Some people would compare it to Phuket before it got developed. We went to Phuket last year and can totally see that being the case.
Phu Quoc has maybe 3 concrete roads, with most of the island being undeveloped and dirt road.
Only recently has there been any real development of the beaches.
If you like to ride around on a scooter, exploring and visiting different beaches, then you'd probably have a great time.
If you're the type who needs modern ameneties...then probably not! Also, probably not the best for singles (nightlife = barely existant).

Were the roads there easy to bike on? I'm a novice biker at best, and my wife has never done it before, so if there's any real traffic then I'd probably stick with paying people to drive me around.

It depends what your definition of "easy" is. My gf and I aren't exactly moto rally racers or whatever, but there are enough automatic bikes around for people like us.
The thing with the roads is that there are rocks sticking out of them, so you do need to stay awake. A friend who we met in Phu Quoc was a fairly experienced rider, but took a bad spill. Just don't go excessively fast, use common sense and you should be fine.
No real traffic except in the city centre. At night or in bad weather, I'd highly recommend paying the locals to drive you. It will only SEEM like you're going to die.

Went to Koh Samet twice; in '01 and '02. It's where the Thais actually go to vacation, and though I've been to Vietnam, I've never been to Phu Quoc.

Samet isn't really a party island; it's more of a hang, though I daresay it has the best food in all of Thailand--the restaurants there were killer, and the beach was pristine. Accomodations were fine, though I went on the cheap and could have paid more.

I'd say go to Samet--it's not as tourist laden as Koh Tao, Phi Phi Don, Phuket, etc. Probably because it's the only option not available by flights, but it's worth the 3 hour drive.

I can't say b/c I've never been to vietnam, but I had a good few days chillaxing at koh samet and would like to return.

I rented a scooter while I was there and had no problems, tho I'd hardly ever driver one before. The main road snaking around the developed part of the island is more or less safe, but the dirt paths off the beaten path can be a little dangerous. Just be safe and drive unimpaired.
