Initially I was planning on going to korea to secure a job in person in an attempt to find an ideal position. However, it has come to my knowledge that this might not be possible anymore due to changes in visa regulations. Is it the case that since I am a first time teacher I must get my E2 visa from my home country(America) and would not be able to do a visa run to Japan, etc. Is it correct that as a first time teacher I must agree to a contract from abroad in order to teach in korea?

No. You could always come to K-land on a tourist visa, get a job and fly/take a ship back to your home country to get a work visa; then come back to start work.

It is possible to do a visa run to Japan for your first visa as long as you have your education verified by the KCUE.

ytuque is correct (assuming you have no record of past Immigration 'offences' i.e. overstaying a tourist visa). If you're never been to K-land, and have all your required documents, it's possible to do a visa run to a country other than your home.

Yep, no need to go all the way home. However, be aware that it can take up to two weeks for the degree verification, and the process must be initiated by your future employer. Many do not know or wish to know how to go about this process.
