"Down cold" means to know something perfectly. For a routine, like a dance routine, it would mean that you could perform (or teach) the routine without having to think about what comes next. It is second nature to you.

In your sentence, it sounds like "she" is a coach or a director. It means that she rehearses her cast often enough and well enough that they perform flawlessly.
