I read a sentence:

Mother's Day is on the second Sunday in MAy.

Q: Is the preposition, in, right?

Isn't it of? -->Mother's Day is on the second Sunday of MAy.

of May sounds accurate.
in May isn't grossly wrong either.

Both are correct. In the article "Mother's Days in various parts of the world" Wikepedia, http://en.wikipedia.org/wiki/Mother's_Day, uses the prepostion "in".
